Adapalene is a second-generation topical retinoid primarily used in the treatment of mild-moderate acne, and is also used
off-label to treat keratosis pilaris as well as other skin conditions. It is effective against acne conditions where comedones are predominant.

Adapalene, also known as 6 - [3 - (1-adamantyl) - 4-methoxyphenyl] - 2-naphthoic acid, has a molecular formula of c28h28o3. It is white or almost white powder at room temperature, insoluble in water or ethanol, slightly soluble . Adapalene is used in dermatology. It is suitable for the skin treatment of acne vulgaris with acne, papule and pustule as the main manifestations. It can also be used to treat acne on the face, chest and back.

 

Adapalene is a retinoid, in vivo and in vitro models of inflammation were demonstrated to have anti-inflammatory properties.Stable chemical structure of adapalene, in the air and light without decomposing. Mechanism and adapalene with specific nuclear retinoic acid receptor combined with retinoic acid is the combination of adapalene and protein and binding of cytoplasmic receptors.

In mice to establish animal model of skin drug test proved that adapalene can treat acne, and acne vulgaris and differentiation process as a role. The mechanism of adapalene is considered by the hair follicle epithelium of normal cell differentiation and reduce microcomedo formation.

Function

1.Adapalene, a new retinol drug, is a Naphthalic acid derivative of all aromatic acids, belonging to the third generation of retinoids. Like other topical retinoids (RA), adapalene has a wide range of pharmacological activities, including anti-inflammatory, anti proliferative and so on

 

2.Adapalene can regulate the differentiation of normal human keratinocytes. Adapalene was 3 times stronger than RA in inhibiting the differentiation of F9 cells. In addition, adapalene increased the production of neuraminidase by keratinocytes, thus increasing the barrier function within the infundibulum of hair follicles.

 

3.Adapalene has a strong inhibitory effect on the proliferation of HeLa cells. The differentiated cells from cervical cancer have high proliferative activity and have been routinely used to study the antiproliferative effects of retinoids.

 

4.Adapalene inhibits the oxidative metabolism of arachidonic acid mainly by inhibiting 5-lipoxygenase and 15 lipoxygenase pathway in human polymorphonuclear leukocytes, thus inhibiting the chemotaxis of human peripheral blood neutrophils.

 

5.Adapalene has unique chemical stability, high stability to light and oxidants, and high lipophilicity, which is easy to pass through hair follicles.

 

6.Adapalene is a kind of microcrystalline suspension in structure. The particle diameter of adapalene is 3-10 ixm, which makes the drug more residual in epidermis, less absorption in dermis, and lower systemic side effects.

 

7.There are many reports about the topical use of adapalene in the treatment of other keratosis and pigmentation disorders, including peritrichoderma, Darier's disease, verruca plana, melanosis of hair follicle, solar keratosis and some light damaged skin diseases.
